Homocysteine Inhibits Hydrogen Peroxide Breakdown by Catalase
نویسنده
چکیده
Catalase, an antioxidant enzyme responsible for degradation of hydrogen peroxide, is protective in many diseases. The amino-acid homocysteine has been suggested to be a pro-oxidant with elevated levels linked to the oxidative stress seen in Alzheimer’s and other diseases. This study shows that homocysteine inhibits the breakdown of hydrogen peroxide by catalase. Physiological concentrations of homocysteine inhibited catalase breakdown of hydrogen peroxide. The inhibition of catalase was by generation of the inactive catalase compound II and prevented by ethanol or NADPH but unaffected by iron. Physiological concentrations of homocysteine also inhibited cellular catalase but were not cytotoxic. homocysteine enhanced the toxicity of the amyloid-ß peptide in an in vitro model of Alzheimer’s neurodegeneration. The antioxidant vitamin E blocked the toxicity of amyloid-ß plus homocysteine but not the catalase inhibition. Catalase inhibition by homocysteine may increase the levels of hydrogen peroxide and play a role in the pathology of disease.

Endothelial cell injury due to copper-catalyzed hydrogen peroxide generation from homocysteine.
We have examined whether the toxic effects of homocysteine on cultured endothelial cells could result from the formation and action of hydrogen peroxide. In initial experiments with a cell-free system, micromolar amounts of copper were found to catalyze an oxygen-dependent oxidation of homocysteine. The molar ratio of homocysteine oxidized to oxygen consumed was approximately 4.0, which suggest...
متن کاملHomocysteine, a thrombogenic agent, suppresses anticoagulant heparan sulfate expression in cultured porcine aortic endothelial cells.
Previous studies showed that homocysteine, a thrombo-atherogenic and atherogenic agent, inhibits an endothelial thrombomodulin-protein C anticoagulant pathway. We examined whether homocysteine might affect another endothelial anticoagulant mechanism; i.e., heparin-like glycosaminoglycan-antithrombin III interactions. Incubations of porcine aortic endothelial cell cultures with homocysteine redu...
متن کاملA spectrophotometric method for measuring the breakdown of hydrogen peroxide by catalase.
Several methods have been developed for following the breakdown of hydrogen peroxide catalyzed by catalase, but these either have not been sufficiently quantitative or have not proved rapid enough to yield reliable data during the critical 1st or 2nd minute of the reaction. Chemical procedures in which residual peroxide is titrated with permanganate (l-3) or an excess of permanganate is measure...
متن کاملEffects of cysteine on growth, protease production, and catalase activity of Pseudomonas fluorescens.
Cysteine inhibits growth of and protease production by Pseudomonas fluorescens NC3. Catalase activity in P. fluorescens NC3 was increased by cysteine. The addition of exogenous hydrogen peroxide did not increase catalase activity, thus suggesting a role for the endogenous generation of hydrogen peroxide via the autoxidation of cysteine.
متن کاملHomocysteine increases nitric oxide synthesis in cytokine-stimulated vascular smooth muscle cells.
BACKGROUND Elevated plasma homocysteine levels have been reported to be an independent risk factor for vascular disease. However, there have been no reports concerning the effects of homocysteine on the production of nitric oxide (NO), another modulator of vascular function and proliferation, by the vascular smooth muscle. METHODS AND RESULTS We investigated the effects of homocysteine on NO ...
